Breast cancer cell lines of different origin reveal different doubling times and p53 functionality. (A) Breast cancer cell lines used in the present study. Molecular subtype, tumor type and receptor status have previously been reported [29], and p53 mutational status was determined by cDNA sequencing. (B) Growth curves of breast cancer cell lines were generated in the presence of 2.5% or 10% FCS, respectively. Means ± s.d. of triplicates are shown. (C) Doubling times of exponentially growing cells were calculated from the data shown in (B). (D) p21WAF1 induction as an indicator of p53 functionality was examined in whole cell lysates after irradiation with 4 Gy at the indicated time points by 6-15% SDS-PAGE (300 μg protein extract per lane) and subsequent immunoblot analysis. Vinculin served as a loading control.
